OBJECTIVE
This experiment provides knowledge of the general characteristics of the genus Streptococcus for the differentiation of species belonging to this group. Additionally, the experiment develops the ability to recognize bacteria of the genus Streptococcus isolated in biological samples in the clinical laboratory, from visualizing the colony in the initial culture to identifying the microorganism. As part of the activities, you will have to learn about the functioning of biochemical tests used in the routine of the clinical laboratory, in addition to learning how to report the result and resolve probable changes in biochemical tests.

At the end of this experiment, you should be able to:

Identify morphologically macro and microscopically Streptococcus spp.;

perform differential tests for other Gram positive cocci;

perform differential tests for different species.

2. Where to use these concepts?
Knowing how to identify bacteria belonging to the genus Streptococcus is a prerequisite for developing experimental skills and abilities that enable the diagnosis of infections caused by these microorganisms. Furthermore, correct identification enables quick and appropriate treatment for affected individuals.

3. The experiment
In this experiment, Streptococcus spp. will be identified morphologically macro and microscopically. For this, various inputs will be used, such as: countertop disinfection kit (alcohol and hypochlorite), Gram dye kit (crystal violet, lugol, ethyl alcohol, fuchsin or Safranine), physiological solution (saline 0, 9%), immersion oil, 3% hydrogen peroxide, bacitracin discs, trimethoprim-sulfamethoxazole discs, optochin discs, PYR test, hyperchlorinated broth, Camp test, bile esculin, bile solubility test, blood agar of 5% sheep containing species of the genus Streptococcus α, β, δ hemolytics and instruments that will serve to assist in carrying out the practice, such as slides, Pasteur pipette (if the dye bottle does not have a dispenser), demographic pencil, lamp and microscope .

4. Security
In this practice, gloves, a mask and a coat, also called a dust jacket, will be used. Although the practice does not pose a risk to the student, these three protective equipment are essential for the laboratory environment. The glove will prevent any possible cuts or contamination with agents harmful to the skin, the mask protects against possible aerosols and the lab coat protects the body as a whole.

5. Scenario
The experiment environment features a Bunsen burner positioned on the workbench, as well as supplies and instruments. You must select and use them to ensure the correct execution of the experiments.
